自由基、细胞凋亡与衰老关系的研究进展
病理改变是脑血管壁β-淀粉样蛋白变性,神经原数量减少,神经纤维缠结。有人认为AD的病理变化是细胞凋亡过盛而导致神经原退行性变〔14〕。Benzi等〔15〕报道了氧自由基在衰老和AD中的重要作用,指出脑组织中含高浓度不饱和脂肪酸、儿茶酚胺和高水平的氧化代谢,是氧自由基最易侵袭的靶器官,而脑内保护性抗氧化酶和非酶性自由基清除剂含量却很低,同时发现AD患者脑的神经纤维缠结中有血红素氧化酶-1、Cu-Zn-SOD,说明氧自由基与神经纤维缠结形成有关,表明氧自由参与了AD的病理过程。推测氧自由基是通过细胞凋亡而加重AD的病理变化。有人证实自由基促进β-淀粉样肽的聚积〔16〕,同样β-淀粉样肽可诱导神经原细胞凋亡〔17〕。详细机制有待探讨。Chen等〔18〕曾经证明细胞早期立即反应基因的表达及产物的活性是有氧化应激控制而且能与神经原内的DNA修复系统相互作用。所以可以推测,β-淀粉样蛋白可诱导氧自由基的产生,氧自由基通过触发细胞早期立即反应基因而引起细胞凋亡。AD与氧化应激密切相关,氧自由基亦可直接引细胞凋亡导致神经原死亡。
